Thymidine kinase 2 deficiency (TK2d) is a sporadic genetic disorder that results in mitochondrial dysfunction, leading to inadequate energy production in cells. It was first diagnosed in children in 2001 whereas improved genetic testing led to the discovery of an
adult form of TK2d in 2013. Common symptoms of TK2d are muscle weakness that gets worse over time, low muscle tone (hypotonia, also called floppy baby syndrome), trouble breathing, problems chewing and swallowing, loss of motor skills (like crawling, walking, balancing, and grabbing), poor reflexes, neurological effects, such as seizures or altered brain activity and function, slowed mental development, hearing loss, ptosis, inability to move the eyes and eyebrows, etc.

Report Key facts:-

1. As per study by Domínguez-González,C.et al. (2019), The late-onset is the less frequent form of presentation of the TK2 deficiency and its natural history is not well known. Patients with late onset TK2 deficiency have a consistent and recognizable clinical phenotype and a poor prognosis, due to the high risk of early and progressive respiratory insufficiency.

2. As per the study titled “TK2-Related Mitochondrial DNA Maintenance Defect, Myopathic Form”, conducted by Wang et al. (2018), the disorder appears to be rare, with approximately 107 affected individuals with a molecularly confirmed diagnosis.

“As per the study titled “Retrospective natural history of thymidine kinase 2 deficiency”, conducted by Garone et al. (2017), 92 patients with genetically confirmed diagnoses of TK2 deficiency were identified in which 67 were from the literature review and 25 were from unreported cases”
